If a Revit template file is open by a user, other users will be unable to create new project files using this template. They will receive the following dialog box after attempting to create a new project:
Say User1 has the default-test.rte template file open and is making some changes. User2 attempts to create a new project using this template through R > New > Project. They browse to the template or have it set as the default. They will receive the dialog box afterward, and it will not go away until User1 is out of the template.
It is recommended that when changes are being made to a template file, stored in a central location, that edits are done locally. This way you can avoid having users attempting to access the template file while changes are being made. Just make a copy of the template file on the local workstation, and when complete copy it back to the server location.
Related to this issue, you can check the owner of a file through Windows should you need to verify the last user to save the template file. In Windows Explorer simply add the Owner column to see the Windows user name associated with the file. Right Click > on the top bar and check Owner:
